α-Chymotrypsin, which contains several NH2 substituents, can be immobilized on an ethylene-maleic anhydride copolymer.
Â
a) Show by reaction scheme the synthesis of immobilized α-Chymotrypsin.
Â
b) The enzymatic activity of the immobilized enzyme shows a maximum at pH=9.5, compared to pH=8 for free α-Chymotrypsin. Addition of NaCl to the immobilized enzyme causes the maximum activity to shift back to pH=8. Explain why.
