lactate dehydrogenase (LD)

pyruvate + NADH -> [S] Lactate +NAD+ is catalysed by lactate dehydrogenase (LD)

 

The UV spectra(using 1cm cuvettes) of 40μM solutions of lactate, pyruvate, NAD+ and NADH are shown below in Fig 1.

 

What is the e340 of NADH? what is the e280 of NAD+?

 

Amixture of excess lactate and NAD+ were placed in a 1cm cuvette and lactate dehydrogenase (LD) solution (this would be a serum in a clinical chemistry lab) added at t=0, Fig2 shows the plot of absobance at 340nm with time. Calculate the rate of this reaction in fig1.pngfig2.png/L/min. Show your calculation.
